You're correct, the weather in Yellowstone is highly variable.
I have been in the park each September for the last six years,
and I would say that the chances of a snowfall during your stay
is a definite possibility. I've never encountered much rain in
the Fall, but if you have some lightweight raingear, it might not
be a bad idea to bring it along. Daytime temperatures can vary from
comfortable 70s down to rather chilly 30s and 40s.
As for the wildlife, well September is an amazing time to be in the park.
In my opinion, the elk are the primary attraction as this is the start
of the rut. Some really good spots to see them are in and around
Mammoth, at Norris and Gibbon Meadows and at ElK Park, as well as
along the Madison River Valley.
Lots of other animals to see as well. Trumpeter Swans usually put on a
great show on the Madison River. Look for moose at Willow Park and
between Norris and Canyon junctions. If you are interested in wolf watching
then Lamar is a good spot to hang out in the early mornings and later
on in the evenings.
You might also be able to see some River Otters in Hayden Valley - they have
been highly visible over the couple of years. Should be lots of bison in
Hayden Valley, and there are usually several blacktail deer hanging out in
the meadows just south of Canyon.
Sorry, but I don't have any information about fishing.... hopefully somebody
else will be able to help you out with that one!
